Abstract  
This study aims to assess the in vivo immunological effect of the ps 80 when it was tested alone, or when the immune system was challenged with Pseudomonas aeruginosa antigen.
 
The study 80 involved random grouping of Albino Swiss male rats into four groups. Each group consisted of three rats. Group 1, injected with seven doses of somatic antigen Pseudomonas aeruginosa.
 
Group 2, injected with six doses of polysorbate 80 of 6% (v/v). Group 3 injected with seven doses of combinations of antigen and surfactant in a volume ratio of 1:1. Group 4 injected with distilled water and considered as a control group. Blood samples were collected from both control and tested groups to measure phagocytic index and interferon-?.
 
The results revealed that when polysorbate 80 was injected in combination with Pseudomonas aeruginosa antigens, produced a highly significant (p< 0.01) enhancement of phagocytic index but it showed no significant changes in the IFN-? level when compared with those challenged with the antigen alone.
 
As a conclusion, polysorbate 80 showed an adjuvant activity when combined with bacterial antigens, represented by the elevation of the level of phagocytic index but not IFN-? level.
 
Introduction   

 In immunological technologies, administration of many vaccines by their own stimulates a poor host immunological response.
 
 This is particularly true of the more recently developed subunit vaccines, thus a group of substances called immune adjuvant are frequently applied. An adjuvant defined as any material that enhances the cellular and/or humeral immune response to an antigen (10).
 
Adjuvant generally elicits an earlier, more potent and longer lasting, immunological reaction against co-administered antigen. In addition, the use of adjutants can often facilitate administration of reduced quantities of antigen to achieve an adequate immunological response (11).
 Surfactants are of great rule especially nonionic surfactants as immune adjuvant to reduce the amount of vaccine to be injected, to increase the immunogenicity of some other added conjugates like bovine serum albumin as well as nonionic block co-polymers (NBPs).
 
It was found that NBPs with hydrophilic POE (polyoxy-ethylene) ends manifested greater adjuvant activity than NBPs with hydrophobic POP (polyoxypropylene) ends.
 
 Several surface-active agents able to stimulate the humeral immune response block polymers with POP chains flanking a POE center, as well as polymers with POE chains flanking a POP center, displayed high adjuvant activity.
 
 These block polymers stimulated the antibody response in a dose-dependent manner. They stimulated the antibody response with both high and low antigen doses. Nonionic block polymer surfactants are potent adjuvant for stimulation of the antibody response (14).
 
Animals, Materials and methods
 
Animals:

Albino Swiss male rats brought from the Embryo Research and Infertility Institute- University of Baghdad. They were caged for acclimatization for 3 months in the Animal House- College of Medicine – University of Babylon. Rats’ weight was ranged 150-250 g. The animals were kept on normal pellet diet, standard room temperature and normal diurnal rhythm.
